Route Creation using Google Maps

I am looking forward to automating routes. I need a google maps expert. The scenario is as follows: I have the location information (coordinates) for a couple thousand users with different pick-up and drop-off locations and with different arrival time requirements to their destinations. I also have shuttles with 16 people capacity. I want to fully automate the route creation process for this ride-sharing process. I want to create routes where everybody reaches their destination on time sharing their rides and using our vehicle types (some time (ex. one hour) earlier to exactly on time). I want to fully automate this process using google maps.
